My son married a woman who is self-employed. Her 1099 included reimbursement for mileage. Can I subtract her mileage at .55 on her Schedule C as an expense?

Maybe. You need to see if the vehicle is (or was) being depreciated in the prior year(s), because the 55 cents per mile contains a good component for depreciation, and the IRS frowns on double dipping. In that case, go with actual expenses for the vehicle.
Otherwise, take the 55 cents per business mile for 2009.
