If I have a 2 register license, can I have 2 checkout lanes and a backoffice for database and management reporting? Or do I have to have a three lane license?

Yes. Just to amplify Ozzie's response, Help verifies exactly what he says. From the horses mouth, if you will.
You can view it here: Help -> Index -> type in "Lic" and select Licensing, Microsoft Point of Sale.

Actually the way it works is that you can have as many machines you want on the network.
This license allows you to run the POS program on 2 machines _at the same time_. It doesn't matter which 2. With a 2 lane license, when you attempt to open POS on the 3rd one, it gives you an error message about exceeding the license
