Not without some sort of Add-in. You could do it with a "hook function" in a COM add-in, or with a SQL Trigger.
Probably the safest thing to do would just be to use options in RMS to "prompt for sales rep" and "require sales rep". If you tell your cashiers they aren't getting commission unless they set the sales rep, they'll remember to do it...
